It had intended to sign-on in Summer 1987, but because many of the programs it was to carry would not become available to them until the fall, a September debut was made. WNYB had secured the rights to Star Trek: The Next Generation
, a major first run syndicated program. Due to the uncertainty that the station was going to be on the air by the fall of 1987, Paramount
, the syndicator of the program (and the later buyer of the TVX chain) opted out of the deal with this channel and instead the program premiered on WUTV
. The station's slogan was "Buffalo's Superstation". Fox was not happy with the ratings it was receiving on WUTV and switched affiliation with WNYB in the fall of 1989.
In that year, the station was sold to Act III Broadcasting
which almost immediately turned around and offered to buy WUTV from Citadel Broadcasting
. Citadel accepted the offer in 1989 and the sale was finalized in June 1990. Act III moved WNYB's stronger programming, including its Fox affiliation, to WUTV. WNYB was then sold to Tri-State Christian Television
and began to carry religious programming full-time including programming from the Trinity Broadcasting Network
. Grant Broadcasting acquired analog channel 26 in Jamestown
in 1995 and negotiated with Tri-State Christian
Television for WNYB in exchange for channel 26 and cash as well as a new broadcasting facility.
The station changed hands in the spring of 1996 and became a WB affiliate at night with Kids WB programming airing late in the afternoons. TBN and other Christian programming continued to air for the rest of the day until September 1996. At that point, Trinity Broadcasting continued to air from 9 in the morning until noon, from midnight to 7 a.m. on weekdays, and until 3 p.m. on Sundays. It also adopted the current call sign WNYO-TV. Because channel 26 was still not operable, the Christian WNYB intellectual unit was unable to move there in 1996. Finally in January 1997, the Christian programming moved to channel 26 along with the WNYB
call letters. The Sinclair Broadcast Group purchased WUTV in 1997 and WNYO in 2001 making the two sisters.
On January 24, 2006, The WB and UPN
announced that the networks would end broadcasting and merge. The new combined network would be called The CW. The letters would represent the first initial of its corporate parents: CBS
(the parent company of UPN) and the Warner Bros.

On February 22, News Corporation
announced that it would start up another new network called MyNetworkTV. This new service, which would be a sister to Fox, would be operated by Fox Television Stations and its syndication division Twentieth Television. MyNetworkTV was created in order to give UPN and WB stations, not mentioned as becoming CW affiliates, another option besides becoming Independent
. It was also created to compete against The CW. MyNetworkTV was launched on September 5; WNYO became an affiliate of the network.
WNYO turned-off its analog signal on June 12, 2009 as part of the DTV transition in the United States
. On the same day, the station moved its digital operations from channel 34 to its previous analog position on channel 49. Its pre-transition digital transmitter was located on the northeastern side of Grand Island
and aired on UHF channel 34.
-produced broadcast on WNLO. Known as WB 49 News at 10, it was part of Sinclair's controversial centralized News Central
operation based at company headquarters on Beaver Dam Road in Hunt Valley, Maryland
. Although national news, weather forecasts, and some sports originated from News Central, local news and sports operations were based at WNYO's studios. It also aired "The Point", a one-minute conservative political commentary, that was also controversial and a requirement of all Sinclair-owned stations with newscasts.
The company ultimately chose to add a news operation to this station as opposed to sister station WUTV despite the fact that Fox requested its affiliates to air local news in the early-1990s to strengthen programming on the young fourth network. This was due to the fact that the ratings of the syndicated WUTV rerun sitcoms at 10 p.m. were especially high in southern Ontario. Ironically, WUTV was far more popular in Canada than it was its own home market, at times even beating independent CHCH-TV from Hamilton, Ontario. Despite record growth in ratings by WNYO's broadcast (much higher than the 8 to 10 p.m. WB block could achieve) and a five year verbal commitment to its staff from Sinclair, the station's news department was eventually shutdown. That was due to poor financial planning by Sinclair. It occurred as part of the systematic shutdown of the company's news operations around the country that began roughly twelve months after the inaugural newscast on WNYO. News Central officially folded at the end of March 2006.
On April 13 of that year, Sinclair announced NBC
affiliate WGRZ-TV
(owned by Gannett) would begin producing a new weeknight prime time show at 10 for WNYO through a news share agreement. This would be a second attempt by WGRZ to air a newscast at 10 after producing one on Pax (now Ion) affiliate WPXJ-TV
from 2001 until 2003. Originally to be called 2 On NYO 10 at 10, the WNYO show began airing on April 20 known as 2 News on 49, 10 at 10. The production is currently known as Channel 2 News at 10 and originates from WGRZ's studios on Delaware Avenue/SR 384
in Downtown Buffalo. The newscast originally aired ten minutes of news and weather with a sports show, called The Sports Zone, completing the half hour. However due to low ratings, The Sports Zone was cut to only 6–8 minutes of sports news. It has consistently lagged behind the WIVB-produced show on WNLO in the ratings because it is not a seven day operation.
As of February 17, 2010, WGRZ became the first Buffalo station to offer its newscasts in enhanced definition widescreen
. However, all of the video is still produced entirely in 4:3 standard definition which is then cropped to a 16:9 aspect ratio and upconverted to 1080i
in the control room for broadcast. The WGRZ show on WNYO was not included in the upgrade due to this station's lack of an HD master control facility. As a result, it is still seen in pillarboxed 4:3 standard definition. WGRZ upgraded to full HD on October 29, 2011; however, the WNYO show remains in pillarboxed 4:3 SD for the time being.

. Although national news, weather forecasts, and some sports originated from News Central, local news and sports operations were based at WNYO's studios. It also aired "The Point", a one-minute conservative political commentary, that was also controversial and a requirement of all Sinclair-owned stations with newscasts.
The company ultimately chose to add a news operation to this station as opposed to sister station WUTV despite the fact that Fox requested its affiliates to air local news in the early-1990s to strengthen programming on the young fourth network. This was due to the fact that the ratings of the syndicated WUTV rerun sitcoms at 10 p.m. were especially high in southern Ontario. Ironically, WUTV was far more popular in Canada than it was its own home market, at times even beating independent CHCH-TV from Hamilton, Ontario. Despite record growth in ratings by WNYO's broadcast (much higher than the 8 to 10 p.m. WB block could achieve) and a five year verbal commitment to its staff from Sinclair, the station's news department was eventually shutdown. That was due to poor financial planning by Sinclair. It occurred as part of the systematic shutdown of the company's news operations around the country that began roughly twelve months after the inaugural newscast on WNYO. News Central officially folded at the end of March 2006.

in Downtown Buffalo. The newscast originally aired ten minutes of news and weather with a sports show, called The Sports Zone, completing the half hour. However due to low ratings, The Sports Zone was cut to only 6–8 minutes of sports news. It has consistently lagged behind the WIVB-produced show on WNLO in the ratings because it is not a seven day operation.

As a result, it is still seen in pillarboxed 4:3 standard definition. WGRZ upgraded to full HD on October 29, 2011; however, the WNYO show remains in pillarboxed 4:3 SD for the time being.
