THP1: This product is used to test leukemia
Leukemia
Leukemia or leukaemia is a type of cancer of the blood or bone marrow characterized by an abnormal increase of immature white blood cells called "blasts". Leukemia is a broad term covering a spectrum of diseases...

 cell lines in immunocytochemical
Immunocytochemistry
Immunocytochemistry is a common laboratory technique that uses antibodies that target specific peptides or protein antigens in the cell via specific epitopes. These bound antibodies can then be detected using several different methods. ICC allows researchers to evaluate whether or not cells in a...

analysis of protein-protein interaction, and immunohistochemistry
Immunohistochemistry
Immunohistochemistry or IHC refers to the process of detecting antigens in cells of a tissue section by exploiting the principle of antibodies binding specifically to antigens in biological tissues. IHC takes its name from the roots "immuno," in reference to antibodies used in the procedure, and...

Characteristics

Morphology: large, round, single cells

Derived from the peripheral blood of a 1 year old human male with acute monocytic leukemia. THP-1 cells have Fc and C3b receptors and lack surface and cytoplasmic immunoglobulins. They also produce IL-1. These cells also stain positive for alpha-naphthyl butyrate esterase, produce lysozymes and are phagocytic (both latex beads and sensitized erythrocytes). THP-1 cells can also restore the response of purified T lymphocytes to Con A, show increased CO2 production on phagocytosis and can be differentiated into macrophage-like cells using for example phorbol 12-myristate 13-acetate  (commonly known as PMA).

Growth Information

This cell line can provide continuous culture, grown in suspension; RPMI
Roswell Park Memorial Institute medium
Roswell Park Memorial Institute medium, commonly referred to as RPMI, is a form of medium used in cell culture and tissue culture. It has traditionally been used for growth of Human lymphoid cells...

1640 + 10% FBS + 2mM L-Glutamine. The average doubling time is 35 to 50 hours.
You may also add 1 mM sodium pyruvate, penicillin (100 units/ml) and streptomycin (100 μg/ml) to inhibit bacterial contamination. Maintain cultures between 2-9x105 cells/ml. 37C, 5% CO2. Cells are non-adhesive.

Hazard: Cell line of human origin. There is no evidence for the presence of infectious viruses or toxic products. However, it is recommended that cultures are handled under Biosafety Level 2 containment.
