He was the eldest son of Claudius Amyand, the Surgeon-in-Ordinary to King George II of Great Britain, and his wife Mary Rabache, and was baptised at St James's Church, Piccadilly

In 1748, Amyand married Anna Maria Korteen, daughter of John Abraham Korteen, a German merchant of Hamburg

. They had two daughters and two sons. Amyand died, aged 45, from undisclosed causes, and was buried at Carshalton

 a week later. He was succeeded in the baronetcy by his elder son George
