He made his Tipperary championship debut in the 2009 Munster Championship coming on as a substitute against Cork.He has twice been named Player of the Tournament at the Kilmacud Crokes All-Ireland Sevens and was part of the Portroe team which won the title in 1999. Darren was captain of the Portroe senior team which reached its first North Tipperary senior hurling final in 2009. He kept a clean sheet, but Eire Og, Nenagh scored enough points to win convincingly.

On 5 September 2010, Gleeson was a non-playing substitute as Tipperary won their 26th All Ireland title
