Auramine O
Encyclopedia
Auramine O, also called Basic yellow 2, Pyocatanium aureum, aizen auramine, Pyoktanin Yellow, Canary Yellow, Pyoktanin, or C.I. 41000, is a diarylmethane dye
used as a fluorescent stain. In its pure form, Auramine O appears as yellow needle crystals. It is very soluble in water and soluble in ethanol
.
Auramine O can be used to stain acid-fast
bacteria
(e.g. Mycobacterium
, where it binds to the mycolic acid
in its cell wall
) in a way similar to Ziehl-Neelsen stain
. It can also be used as a fluorescent version of Schiff reagent.
Auramine O can be used together with Rhodamine B
as the Truant auramine-rhodamine stain
for Mycobacterium tuberculosis
. It can be also used as an antiseptic
agent. According to the packaging of Post-it brand notes, Canary Yellow is a trademark of 3m.
Dye
A dye is a colored substance that has an affinity to the substrate to which it is being applied. The dye is generally applied in an aqueous solution, and requires a mordant to improve the fastness of the dye on the fiber....

Ethanol
Ethanol, also called ethyl alcohol, pure alcohol, grain alcohol, or drinking alcohol, is a volatile, flammable, colorless liquid. It is a psychoactive drug and one of the oldest recreational drugs. Best known as the type of alcohol found in alcoholic beverages, it is also used in thermometers, as a...

Acid-fast
Acid-fastness is a physical property of certain bacteria, specifically their resistance to decolorization by acids during staining procedures.Acid-fast organisms are difficult to characterize using standard microbiological techniques Acid-fastness is a physical property of certain bacteria,...

Cell wall
The cell wall is the tough, usually flexible but sometimes fairly rigid layer that surrounds some types of cells. It is located outside the cell membrane and provides these cells with structural support and protection, and also acts as a filtering mechanism. A major function of the cell wall is to...

Ziehl-Neelsen stain
The Ziehl–Neelsen stain, also known as the acid-fast stain, was first described by two German doctors; Franz Ziehl , a bacteriologist and Friedrich Neelsen , a pathologist. It is a special bacteriological stain used to identify acid-fast organisms, mainly Mycobacteria...

Auramine O can be used together with Rhodamine B
Rhodamine B
Rhodamine B is a chemical compound and a dye. It is often used as a tracer dye within water to determine the rate and direction of flow and transport. Rhodamine dyes fluoresce and can thus be detected easily and inexpensively with instruments called fluorometers...

Auramine-rhodamine stain
The auramine-rhodamine stain , also known as the Truant auramine-rhodamine stain, is a histological technique used to visualize acid-fast bacilli using fluorescence microscopy, notably species in the Mycobacterium genus. Acid-fast organisms display a reddish-yellow fluorescence. Although the...
