The 2010 OAFL Division 2 competition was played between May and October 2010 and consisted of eight rounds. With the introduction of new team the Toronto Rebel Dogs (Toronto Downtown Dingos/Toronto Rebels) and the withdrawal of the Toronto Eagles Reserves. Five teams once again competed for the Mike Pyke Cup. The league used the McIntyre final five system in 2010 changing from the final four system used in 2009 OAFL Division 2 season

The Quebec Saints would go on to claim back-to-back premierships. Saints midfielder Darrin Haverhoek picking up the Best on Ground award for the Grand Final. Central Blues player Earl Monero claimed the regular season league Best & Fairest award while Toronto Rebel Dog Paul Tinkler took out the leading goal kicker with 14 goals.
