This largely depends on local fire regulations related to horizonal fire separation and smoke transfer. To answer your question though it is no, not always.

There are automatic shutter systems and other means of separation of the upper space depending on its size and location within the building at its deign including structural and overall MEP. The escalator itself has very little to do with those requirements.
